Need a good site to run league??? anyone??

Does anyone know of a good website to run a 12 man league that also is able to customize scoring and starting line up formats? I want a 12 team league. I would also like an option on starting lineups to be as varied as 1 RB and 5 WR's, or @ Rb's, 2 WR's and 2 TE's, or basic line up. As long as the combo consist of 6 and no more then 2 RB's? Any good sites?? Thanks for any insight to this. I am relatively a new commish and don't want to flop. Thanks and good luck to all.
